본문 Study and Use of the Microscope and the Micrometer Summary In this experiment, we have studied and practiced the use of the microscope and the micrometer. The micrometer is divided into two parts. The one inserted on the ocular is the eyepiece micrometer, and the other one which is set up on the stage is the stage micrometer. By using the relativity of the two lenses, we are able to determine the measuring unit of the eyepiece micrometer at different magnifications. Researchers use this micrometer in order to measure the length and various values of specimens in the microscope. Introduction When using an ocular microscope to observe a specimen, mainly we stick to these following steps; 1. Turn the power on. 2. Set the objective lens with the lowest magnitude in place. 3. Turn the light on and set the brightness on an appropriate level. 4. Position and fix the slide on the stage. 5. Lower the stage to its lowest height using the coarse adjustment knob. 6. Center the specimen while watching the image. 7. Using the coarse and fine adjustment knobs, focus the image coarsely, then clearly. 8. Look through the ocular lens and observe the specimen. After use, note that the stage must be set on the lowest position and the objective lens must be set at lowest magnification power. When it is not being used, be sure to cover the microscope with a blanket or a bag to protect it from dust. When being used, the microscope must be placed on a flat, level, stable surface. While elevating the stage, beware of the working distance so that the stage and the objective lens does not contact. To carry the microscope, hold the arm and the bottom of the microscope with each hands. You must always be careful with the lens and clean it periodically. Total Magnification = (the magnification of eyepiece lens set to 10X) × (the magnification of objective lens; 5X, 10X, 100X) How will we use this micrometer to measure the length of specimens in the microscope?
