Introduction
We want to know about the transformation in E.coli through this experiment. Transformation means the alteration of phenotype by the genetic change from the uptake and incorporation of external genetic material. We can use E.coli because its cell has competence, the ability to take up extracellular DNA from surroundings, so it is possible to give some genetic changes artificially. What we should consider about in this experiment is how to make transformation to E.coli and how we could discern whether transformation occurred or not.
pGLO is a plasmid that has AMPr, araC and GFP gene. If the genes in the plasmid are transcribed, the cell containing the plasmid get new traits such as resistance from ampicillin. Thus, bacteria which does not have pGLO will be eradicated in an ampicillin medium. AraC gene is active only in the condition with arabinose, and araC protein, a transcription factor combining with the promotor, is essential to initiate the transcription of GFP gene. So if the E.coli containing pGLO lies on the arabinose, that colonies will exhibits bright green fluorescence when we expose them to light in the blue to ultraviolet range.
Procedure
Label to discern whether pGLO contains
2. Transfer 250ul CaCl2
3. place ice
4. Pick up a single colony using a loop
5. Immerse a new loop into the plasmid DNA stock tube.
Withdraw a loopful.
Mix the looful into the cell suspension of the +pGLO tube.
